<i>In vivo</i>immune inhibitory role of Ly-6A protein

چکیده

Abstract Ly-6A is a GPI-anchored cell surface protein with reported adhesion and signaling role in CD4 +T cells. shows low expression on naïve T cells; however, its elevated upon activation. Additionally, IFN-γ IL-27 cytokines increase expression, feature observed immune checkpoint inhibitory (ICI) proteins, such as PD-1 CTLA-4. Many reports using lines primary cells show, for ICI inhibiting clonal expansion of antigen receptor-stimulated While the published vitro ex vivo studies provide strong evidence protein, unknown. Using tumor transplantation mouse model, we have found that Ly-6A-deficient mice support slower or no growth B16-F10 melanoma MC38 adenocarcinoma tumors compared to wildtype C57BL/6 control mice. We report injected number activated draining lymph nodes (DLN) day-3 post-injection. Furthermore, significantly proliferation DLN from was day-7, assessed by assays. Transcriptome analyses tissues showed, among others, inducible chemokine genes. CD73 confocal microscopy, lower than tumors. Our results reveal novel potential target cancer immunotherapy, either standalone combinatorial blockade strategy. Supported funds Department Biology, Villanova University (SRG) &amp; Sigma Xi

Inhibition of Ly-6A antigen expression prevents T cell activation

Antisense oligonucleotides complementary to the 5' end of the mRNA encoding the Ly-6A protein were used to block the expression of that protein. Using this approach we could inhibit the expression of Ly-6A by 60-80% in antigen-primed lymph node (LN) T cells as well as in the D10 T cell clone. Inhibition of Ly-6 expression resulted in the inability to restimulate in vitro, antigen-primed T cells...

Structure/function relationship of activating Ly-49D and inhibitory Ly-49G2 NK receptors.

Murine NK cells express Ly-49 family receptors capable of either inhibiting or activating lytic function. The overlapping patterns of expression of the various receptors have complicated their precise biochemical characterization. Here we describe the use of the Jurkat T cell line as the model for the study of Ly-49s. We demonstrate that Ly-49D is capable of delivering activation signals to Jur...

Altered Proliferative Response by T Lymphocytes of Ly-6A (Sca-1) Null Mice

Ly-6A is a murine antigen which is implicated in lymphocyte activation and may be involved in activation of hematopoietic stem cells. Antibody cross-linking studies and antisense experiments have suggested that Ly-6A is a lymphocyte coactivation molecule. To better understand the function of Ly-6A, we used gene targeting to produce Ly-6A null mice which are healthy and have normal numbers and p...

The glycosyl phosphatidylinositol anchor is critical for Ly-6A/E- mediated T cell activation

Ly-6E, a glycosyl phosphatidylinositol (GPI)-anchored murine alloantigen that can activate T cells upon antibody cross-linking, has been converted into an integral membrane protein by gene fusion. This fusion product, designated Ly-6EDb, was characterized in transiently transfected COS cells and demonstrated to be an integral cell surface membrane protein. Furthermore, the fusion antigen can be...
